A set of peptides was taken for targeted validation using targeted proteomic approach of Multiple reaction monitoring (MRM)/selected reaction monitoring (SRM). We have performed targeted experiment for two proteins in female gonad tissue of Labeo rohita. Results were matched with a spectral library for the reliability of the data. TSQ Altis Triple Quadrupole Mass spectrometer (Thermo) coupled to an HPLC system was used for acquiring the data. This data represents the significance of PeptideAtlas in targeted experiments.
For two of the selected proteins in female gonad tissue of Labeo rohita, only those peptides and transitions were selected which matched to the spectral library (of same tissue). Data was acquired against the list of transitions and data was analysed in Skyline.
One µg of peptide sample was loaded onto the column and separated for 10 minutes using a liquid chromatography system consisting of solvent A with 0.1% formic acid (FA) in milliQ water and solvent B with 80% Acetonitrile (ACN) and 0.1% FA. A cycle time of 2 sec was used and column temperature was set to be 45 οC throughout the run. The acquired data was analysed using Skyline daily software (version 20.2.1).
