Hi Panorama people!!!

Wonderful job with AutoQC, one of the best tools I've used for QC in the lab... for more than 25 years!!! (I used to plot Shewhart cotrol chart rules by hand!)

So, here suggestion that might work. Due to the generous real state of large screens, it would be very helpful if you could choose more than one metric (eg Rt and Area) side my side for each target. That would sometimes help quickly identifying and diagnosing problems, as switching from metric to metric for a big list of targets makes it hard to visualize. Of course, I could just split the browser into to screens, but then you have to synchronize scrolling (me?? ?lazy???)

You are doing a great job!!! Keep it up!!!

Thanks for the feedback! Glad to hear AutoQC is working for you.

Just to clarify what you're proposing:

It sounds like you'd like to see separate plots for different metrics, not have them as separate series on the same plot like we do for the precursor/transition areas option, correct?

Do you see this as being similar to what happens when you check the boxes to showing moving range and CUSUM plots when the "small" plot option is selected? Except maybe that the plots stay large and just expand to the right?

Yes, something like the CUSUM plots would work, BUT, displaying different metrics (not type of plots). It would be nice to say, plot on one column RT next to Areas and see if there is any direct correlation between the behaviors for the same targets.

I'm curious how close the existing "Transition & Precursor Areas" option is to what you're looking for. It shows a split y-axis plot with the two different metrics as separate series. It is, of course, contingent on having quantitative transition areas. If you don't have data like that in your folder already, you can see an example here:

In your own folders, you can also configure two metrics that you want to plot together that show up in the metric drop-down just like "Transition & Precursor Areas". Here's the doc page:

You can leverage the existing SQL queries and match them up. In the same example folder I linked above, I did just that to pair the retention time and area metrics together. I'm attaching a screenshot showing the configuration.

Obviously it would be nice to be able to pair metrics together without needing to create a separate configuration for each pair, but let me know how close to the mark this approach comes.

That's very helpful. I think that should help. Maybe having this process more streamlined would help (say, two drop down menus with the metrics you wish to combine). That would just make the process you describe more convenient (just being lazy here!).

As q quick follow up, I tried extracting the pressure traces on our experiment (.raw files acquired on QExactive). I'm following the procedure Define Pressure Trace Metrics as in https://www.labkey.org/Documentation/wiki-page.view?name=premPanoConfigMetrics#define but I dont get any options in the 'Use Trace' field when Add New Trace Metric (see attached)

I cant see the pressure trace either when using Skyline (although the .raw files does contain the pressure trace when I open it using Freestyle).

Just wondering if others have had this same issue. Any tips?

Hi Alex,

First, what version of Skyline are you using? I believe that Skyline 20.1 was the first version to extract these traces. You've likely upgraded to something more recent than that. But if there's a chance the RAW file was added with an old version of Skyline, reimporting with the latest version is worth a shot.

If you're still not seeing it, I recommend uploading a representative RAW file, if possible, to the Skyline support page so that the Skyline team can check it out:

Unfortunately, each raw file variant requires effort, so it's possible you're working with a slightly different setup than they've tested with to date.

Thanks for the file. I've learned that Skyline isn't yet extracting the traces for Thermo raw files, but the development team is working on it now. Watch the release notes for an upcoming Skyline daily version!
